Common issues in cell culture:
Stagnation period: The stage during which cells adapt to culture conditions and do not divide. Cells are usually cultured at the beginningWithin 24 hours of attachment, the total duration of this stage depends on the growth stage and seeding density of the cells used at the beginning of cultivation.
Logarithmic phase: Cells actively divide during this stage, which is the time for evaluating population growth and collecting universal data. In the later stage of the logarithmic phase, overcrowding leads to cell stress before the passage of cellsThe timing of (sub cultivation).
Stable period: as cells reachComplete fusion, cell growth slows down at this stage, with less than 1/10 of cells in the active cell cycle. Cells are most susceptible to injury at this stage, so careful observation is necessary to ensure that cells are passaged before or at the beginning of this stage.
Decline period: a natural component of the cell cycle. At this stage, as cell death becomes dominant, the number of live cell populations decreases.

Selection of Cell Culture Plates:
1) Cell culture plates can be divided into flat bottomed and round bottomed (U-shaped and V-shaped) according to the shape of the bottom;
2) The number of culture wells includes 6, 12, 24, 48, 96, 384, 1536, etc;
3) According to the different materials, there are Terasaki plates and ordinary cell culture plates. The specific selection depends on the type of cultured cells, the required culture volume, and different experimental purposes.
Flat bottom and round bottom(The difference and selection of U-shaped and V-shaped culture plates:
1) Adherent cells are generally cultured on flat bottomed plates.
2) The culture of suspended cells is generally done using V-type.
3) U-shaped culture plates are also commonly used for cultivating suspended cells.
4) V-shaped culture plates are sometimes used for immunological agglutination experiments.

Cell culture environment:
Different cells have different culture environments. This is not only about the difference in culture medium, but also about the spatial density of cell growth. Some cells have a larger quantity and better growth status. This usually belongs to cells with slow growth rates, such as endothelial cells. Some cells, such as macrophages and certain tumor cells, grow better when their number is smaller. Especially macrophages, they grow quickly and adhere to the wall quickly, so when passaging, a small amount of cells should be retained so that the cell state is better. And macrophages prefer to grow in clusters, and there is space between clusters. If it grows into a connected whole, the cell morphology will basically deteriorate, and there will be more aging, which is not good for the later experimental results. So when conducting cell culture, it is necessary to explore more related issues such as the cell's preferred growth space density and growth pattern in order to cultivate ideal cells.
