Notes:
1. Sample addition:
① When there are too many experimental samples, they can be operated in batches to reduce errors caused by prolonged experimental time;
② After adding enzyme reagents, use absorbent paper to absorb the reagents outside the well;
③ When conducting quantitative experiments, use a sampler to add reagents and regularly calibrate their accuracy. In addition, ensure one pipette head per sample to prevent cross contamination.
2. Incubation:
① If there are two temperatures, try to use lower incubation temperature and longer reaction time conditions;
② Place a small thermometer in the plate hole reaction solution for measurement and observation;
③ The thermodynamic gradient between the surrounding holes and the central hole of the 96 well plate during incubation can cause "edge effects", so it is recommended to use a water bath as much as possible;
3. Board washing:
① When manually washing the board, it should be perpendicular to avoid cross contamination, and excessive force should be applied to prevent the detachment of antigen antibody complexes;
② When washing the board with a washing machine, it is necessary to regularly check whether the washing head is unobstructed. If it is blocked by debris, a syringe needle can be used to remove it;
③ In order to achieve good washing results, the laboratory can use a combination of machine and manual washing methods. After machine washing, the plates can be manually washed 1-2 times, or the number of plate washes can be appropriately increased;
4. Color rendering:
In the ELISA kit, if tetramethyl (TMB) is used as the substrate, two bottles of application solution A and B are provided as the substrates; If OPD is used as the substrate, the reagent kit can extract tablets or powders. The color reaction conditions are 37 ℃ or room temperature for 15-30 minutes. Reagents based on OPD must be prepared 30 minutes before use and must be kept away from light. Reagents based on tetramethylamine (TMB) do not need to avoid light, but solutions A and B should try to avoid contact with metal instruments as much as possible.
5. Judgment:
The reading of results should be completed within 15-30 minutes. For qualitative determination, when interpreting the test results with the naked eye, the reaction plate should be horizontally 10-15 cm away from the white background. For quantitative determination, the results should be read using an enzyme-linked immunosorbent assay (ELISA) reader, which should not be placed under sunlight or strong light and should be preheated for 15-30 minutes before testing.
