Product Introduction

The PAGE color rapid gel preparation kit (10%) uses the formula in the form of premix, and only needs to add improved coagulant when preparing the gel, which is convenient and fast. The upper layer is colored (purple) gel, which is convenient for sampling. The gel prepared by this product can also be used for non denaturing PAGE gel electrophoresis experiment. This kit is equipped with an improved coagulant, which has no TEMED pungent odor, good stability and catalytic effect. It only takes about 20 minutes to prepare one or more gel. For ease of access, the opened improved coagulant can be stored at 4 ℃ for at least 3 months. Glue quantity (125 gels): 125 pieces (0.75 mm); >90 pieces (1.00 mm); >60 pieces (1.50 mm)

Experimental procedures:

1. According to the thickness of gel prepared as required, take equal volumeLower layer adhesive solutionandlow gel bufferMix well; Add the corresponding volume of improved coagulant and mix well.

2. Immediately inject the prepared lower layer adhesive into the glass plate, so that the distance between the liquid surface and the short glass plate edge is 0.5 cm longer than the comb teeth. Add an appropriate amount of water or anhydrous ethanol to seal the lower layer adhesive and wait for it to solidify for about 10-15 minutes (when a clear boundary line is visible between the sealing solution and the adhesive, it indicates that the adhesive has solidified). Then pour out the upper layer solution. Note: 1) If the volume of solution preparation is excessive, a little can be left in the tube to observe the gel state; 2) When the room temperature is low, the solidification speed is slow, and the gel time needs to be properly extended.

3. According to the thickness of gel prepared as required, take equal volumeUpper layer adhesive solutionandColor upper layer adhesive buffer solutionMix well; Add the corresponding volume of improved coagulant and mix well.

4. Immediately inject the prepared upper layer adhesive into the glass plate, insert the comb, and wait for the upper layer adhesive to solidify (5-10 minutes) before pulling out the comb.

Note:1)Try to use freshly prepared electrophoresis buffer as much as possibleRecommended electrophoresis conditions: 150V for about 50 minutes or 200V for about 35 minutes.

Notes:

1. The upper layer of gel prepared by this product has no concentration effect on the sample, which is similar to the preformed rubber and is similar to the traditionalPAGECompared to glue, protein band separation is more effective, especially for small molecule proteins such as10 kDa)It can also be clearly separated, and the protein bands are narrower and sharper.

2. The dosage of improved coagulant can be adjusted according to personal habits and experience. Adding more coagulants can accelerate gel, and vice versa. Under the same conditions, the higher the temperature is, the faster the gel speed is. If the temperature is too high, it is recommended to reduce the dosage of coagulant properly.

3. Before use, the gel solution and buffer solution can be restored to room temperature, which can effectively avoid the generation of bubbles in the gel process.
