The layout design scheme of PCR laboratory should follow the following principles:

1、 Overall layout of the laboratory

1. Clear zoning: The laboratory should be divided into at least three independent areas, namely the sample preparation area, PCR reaction mixing area, and product analysis area. Each area should have clear physical isolation to prevent cross contamination.

2. Unidirectional process: Personnel and sample flow within the laboratory should follow the principle of one-way process, from the sample preparation area to the product analysis area, to avoid backflow.

3. Air pressure gradient: A certain air pressure gradient should be maintained between each area, usually positive pressure in the sample preparation area, negative pressure in the PCR reaction mixing area, and positive pressure in the product analysis area.

4. Air filtration system: The laboratory should be equipped with a high-efficiency particulate air (HEPA) filtration system to ensure that the air cleanliness meets the requirements of PCR experiments.

5. Experimental platform and fume hood: The experimental platform should be equipped with a fume hood to reduce the diffusion of aerosols generated during the experimental process. The fume hood should have a good exhaust system.

6. Safety measures: Laboratories should be equipped with biosafety cabinets for handling samples that may contain harmful substances. At the same time, there should be emergency shower and eye wash facilities.

7. Cleaning and Disinfection: The laboratory should have clear cleaning and disinfection procedures, and regularly disinfect the laboratory countertops, equipment, and floors.

2、 Decoration materials and facilities

3. Ceiling: Use dust-proof and easy to clean materials, such as aluminum ceilings.

4. Lighting: Provide sufficient natural and artificial lighting to ensure the accuracy of experimental operations.

5. Ventilation system: Design an independent ventilation system to ensure air circulation and pressure gradient inside the laboratory.
