1L Media Protocol
Prepare a nutrient mixture using a 2:1 IMDM culture medium

1. Thaw fetal bovine serum and Pennsylvania/Streptococcus at 37 degrees Celsius

2. Add 626ml IMDM and 313ml nutrient mixture to a 1L filter bottle

3. Add 50 milliliters of FCS

4. Add 10ml of Penn flow

5. Filter

6. Add 1ml of 5mg/ml insulin (or 500ul, 10mg/ml)

Note:Dilute 50mg insulin powder with 10ml sterile H20, add 50ul sterile 1N hydrochloric acid, and store in 4C foil

7. Add 5ug EGF

Note:Dilute EGF make 1ug/ul stock solution with serum-free IMDM, adding 5ul per liter. Store aliquots at -80 ° C.
