Tongwei Biological Reagent KitIt is a highly sensitive, specific, and reproducible experimental diagnostic method. Due to its stable reagents, easy storage, simple operation, and objective result judgment, it has been widely used in various fields of immunological testing. The ELISA detection kit is suitable for in vitro qualitative detection of anti human hepatitis E (HEV) in human serum or plasma.

Preparation of quality control serum:
1. Collect fresh positive serum without hemolysis, jaundice, or bacterial contamination.
2. Heat at 56 ℃ for 10 hours to activate.
3. Centrifuge or filter to remove sediment.
4. Dilute the collected serum with 10% calf serum or normal human serum (PBS buffer) to the desired concentration.
5. Filter and sterilize, divide into small ampoules according to the amount used once, seal, store at 20 ℃ for later use, do not freeze or thaw repeatedly, and the level required to detect the test substance is often considered the level that should be selected for quality control serum.
6. Calibrate the content, delete the mean value of>± 2SD data from 20-30 measurements as the target value, and compare it with the known fixed value serum for measurement.

Preparation method during operation:
Low value samples:Dilute the test sample (including the analyte) with a mixture of human serum (with a lower concentration level of the analyte) or 5% bovine serum albumin physiological saline solution to produce a sample that is close to the lower limit concentration level of the linear range of the method. Generally, there are 5 concentration levels, and the concentration level interval should be less than 10% of the lower limit of the linear range.
High value samples:
1）Select high-value samples containing the analyte, and if necessary, add pure samples of the analyte and calculate the theoretical value.
2）Dilute the high-value test sample with mixed serum or 5% bovine serum albumin physiological saline solution, or the dilution solution required by the measurement method, to approach the upper one-third of the linear range, and record the dilution factor.
3）At least three high concentration samples should be selected, and the dilution factor should be the large dilution factor indicated by the method performance, and the dilution ratio should be appropriately increased or decreased.
