Basic Introduction:


Poly lysine is a positively charged amino acid polymer that can bind to DNA, red blood cell membrane, or any negatively charged protein. It is a non-specific cell adhesion factor that promotes cell adsorption onto a solid matrix. The mechanism of action is that it can enhance the electrostatic interaction between negatively charged ions on the cell membrane surface and solid matrix surfaces (such as cell culture dishes, glass slides, etc.). After adsorption onto the culture surface, polylysine increases the number of cation binding sites used for cell binding. The molecular weight of polylysine is related to viscosity, meaning that lower molecular weight and viscosity make it easier to use; Higher molecular weight, higher viscosity, and more adhesive sites provided. Polylysine with a molecular weight greater than 30000 is suitable for promoting cell adhesion to solid matrices.


Poly lysine has two common subtypes, D - and L-type. Due to the fact that polylysine is a non-specific adhesion factor that binds to cells, both subtypes can be used as solid substrates for coating and promote cell adhesion growth in cell culture. According to reports, poly (L-lysine) can improve the adhesion of proteins on ELISA culture plates. However, in cellular applications, certain cells are capable of hydrolyzing poly L-lysine. In this case, poly D-lysine must be used as an adhesion factor to protect cells from damage caused by excessive uptake of L-lysine.


This product is Poly-D-lysine with a molecular weight of 150000-300000, suitable for cell culture. As a cell culture substrate, the recommended dosage is approximately 0.5ml-1.0ml of 0.1mg/ml polylysine solution for a 25cm2 culture plate. This product can also be used as a sticky tablet for histological analysis.

Preservation and transportation methods

Storage: Dry storage at -20 ° C, stable for 3 years.

Precautions

In certain cell culture applications, some cells digest and absorb poly (L-lysine), and excessive intake of poly (L-lysine) can cause certain cytotoxicity. Therefore, in such situations, it is recommended to use Poly-D-lysine, such as Poly-D-lysine (Mw 150,000-300,000)(#MX0922)。

2. This product is provided in the form of hydrobromic acid (HBr). If you want to remove hydrobromic acid from this product, you can dissolve it in neutral buffer and dialyze to remove salt ions.

Instructions for use

1. Preparation of storage solution

Dissolve 10mg of powder in 2ml of sterile deionized water and filter it through a 0.2 μ m low protein adsorption membrane to obtain 5mg/ml poly-D-lysine storage solution, which can be directly used for cell culture purposes. To maintain contamination caused by repeated use, the storage solution can be divided into sterile tubes in single use quantities and stored at 4 ℃ or -20 ℃ for at least 1 year.


2. Cell culture coating matrix

The use of poly D-lysine for cell related coating experiments requires optimization of coating conditions based on different cell lines and applications. The general steps are as follows:
1) Dilute 5mg/ml poly-D-lysine storage solution with sterile tissue culture grade water or PBS to the most suitable coating concentration for your own experimental system. Normally, the commonly used culture dish/plate coating concentration is 0.1mg/ml, which means diluting the storage solution 50 times before use.

2) Under sterile conditions, coat the cell culture plate with a dosage of 0.5ml-1.0ml/25cm2. Gently shake the bottom of the board to cover the entire surface.

3) After 5 minutes, remove the surface solution of the culture plate with a gun and clean the plate surface with sterile tissue culture grade water or PBS; 【 Attention 】: In some cases, it may take 1-2 hours to complete the board laying, and in other cases, overnight board laying is required. It depends on the situation.

4) After drying for at least 2 hours, cell culture can be carried out.

[Note]: If the glass container or slide coated with polylysine must be sterilized, it can be sterilized by gamma ray irradiation (rather than high-pressure sterilization).

【 Attention 】: If the surface of the coating is uneven, 1mM magnesium acetate can be used to pretreat the glass slide for 2-3 hours, thoroughly clean it, and then start coating. Alternatively, glass slides can be washed with acid (hydrochloric acid or sulfuric acid) to achieve a smooth coating of the polylysine solution.


3. Histological research (coating of glass slides)

It is generally recommended to use a 0.1% (w/v) poly-D-lysine solution for the preparation of histological slides. After treating the glass slide with a corresponding concentration of polylysine solution for 5 minutes, clean the slide and dry it at room temperature overnight or in an oven at about 60 ° C for 1 hour. [Note]: This working solution is stored in a plastic bottle at 4 ° C and is limited to 4 uses.
