Product description

Yeast deficient culture medium is a complete culture medium made by mixing SD Base+DO Supplement with or without agar. The former is a solid medium and the latter is a liquid medium (broth) used for selecting yeast containing specific carriers for cultivation or conducting heterozygous screening.

Minimal SD Base， The Minimum Synthetic Defined Medium is a culture medium foundation synthesized by adding a single carbon source such as glucose, galactose, raffinose, and succinate or a combination of multiple carbon sources to the amino acid free yeast nitrogen source foundation (YNB). Dropout (DO) Supplement， Amino acid deficiency mixture (or additive) is a mixture lacking one or more specific amino acids.

The commonly used SD series culture medium, such as SD/- Trp, is a yeast deficient medium that uses glucose as a carbon source and lacks tryptophan (Trp). Under the premise of keeping other components unchanged and replacing different carbon sources, various combinations of yeast defect type culture media are produced, such as SG/- Trp (using galactose as the carbon source), SF/- Trp (using fructose as the carbon source), SM/- Trp (using maltose as the carbon source), SS/- Trp (using sucrose as the carbon source), SR/- Trp (using raffinose as the carbon source), etc., to meet different research scenarios.

SG/- Ura with Agar is a yeast deficient solid culture medium that uses galactose as a carbon source and lacks uridine (Ura). It is mainly used for yeast expression and screening experiments that require galactose induction.

Preservation and transportation methods

Storage: Dry storage at+4 ℃, stable for at least 2 years.

Transportation: Room temperature transportation.

cultivateNutrient base preparation

1) Take a packet of powder, add 500ml of deionized water to it, stir and mix well to dissolve it. Agar cannot dissolve until after high-pressure sterilization.

2) Important: Test whether the pH of the culture medium is 5.8, and adjust the pH with weak acid or weak base if necessary.

3) Sterilize under high pressure at 121 ℃ for 15 minutes. Cool to~50 ℃ before flipping the board.

4) At room temperature, allow the agar plate to solidify, seal it with a sealing film, and store it upside down at 4 ℃.

Precautions

1) Users can purchase various yeast defect type culture media or basic raw materials according to their own experimental needs, as shown in the composition diagram of yeast screening culture media below.

2) When preparing SD (including SG, SM, SF) series yeast culture media, the color of the media will change from light yellow to dark brown, and it will not have a serious impact on the growth of yeast.

3) If you want to avoid carbon source carbonization during high-temperature sterilization of the culture medium, you can choose SC medium or YNB+DO supplement. Before the experiment, adding a sterile carbon source separately can also prepare a complete yeast screening medium.
