Experimental operation guide:

Western Blot (WB) procedure

Sample preparation: lyse cells or tissues, extract total protein, quantify and denature.

Electrophoresis: Separate proteins by SDS-PAGE and transfer to PVDF membrane.

Closure: 5% skimmed milk powder is sealed at room temperature for 1 hour.

Incubate antibodies: primary antibody (diluted 1:1000) at 4 ℃ overnight, secondary antibody at room temperature for 1 hour.

Detection: ECL chemiluminescence development, light avoidance operation.

Immunohistochemistry (IHC) steps

Slicing processing: dewaxing hydration antigen repair.

Closure: 3% H ₂ O ₂ inactivates endogenous enzymes, and serum blocks non-specific sites.

Incubate antibodies: primary antibody (diluted 1:100) at room temperature for 1 hour or overnight at 4 ℃.

Color development: DAB color development, hematoxylin counterstaining, dehydration sealing.

Key precautions

Dilution ratio: Dilute strictly according to the recommended ratio. If it is too high, it may cause non-specific binding, and if it is too low, it may reduce sensitivity.

Light avoidance operation: The color development step needs to avoid light to prevent signal attenuation.
